What Is Amplicon Sequencing by Nanopore Technology?

Amplicon Sequencing uses long-read technology to generate full length DNA sequences that have been amplified using PCR between 500 bp to 150 kb. This service is ideal for clonal populations as it delivers a consensus sequence of the sample. Linear and PCR samples are required as starting materials.

Quality Check

Quality score is determined by analysis software. Targeted data size based on the targeting coverage. Any results that do not meet the targeting data size can be translated to insufficient sample DNA. Sequencing results are directly dependent on the quantity, quality, and purity of the sample DNA. 

Sources of Inadequate Sample Quality

Insufficient DNA concentration

  • Verification on Qubit.

Degraded and/or contaminated samples

  • Caused by poor PCR purification technique or reagents/primers remaining in the final product.
  • Caused by multiple freeze/thaw cycles and/or exposure to extreme temperatures.

Multi-species sample

  • This service is intended for single amplicon samples. Assembly and analysis of multi-amplicon sample may fail.
  • Despite producing final results, the final assembly and analysis may not be an accurate representation of a multi-amplicon sample.
